It has been reported that the fruits and leaves of berries such as the blackberry, raspberry, and strawberry contain a high level of scavenging activity for chemically generated active oxygen species. This study investigated the antioxidative activities of black chokeberry fruit (Aronia melanocarpa Elliot) both in vitro and in vivo using the DPPH stable radical and rats with ethanol-induced gastric injury, respectively. The red pigment fraction of the black chokeberry contained three main components, one of which was identified as cyanidin 3-O-beta-glucoside by HPLC analysis and (1)H NMR. The black chokeberry red pigment fraction scavenged >44% of DPPH radicals at a concentration of 25 microg/mL compared to the control solution. The black chokeberry extract and its hydrolysate administrated at 2 g/kg of body weight each had nearly the same protective effect as quercetin administrated at 100 mg/kg of body weight in suppressing the area of gastric mucosal damage caused by the subsequent application of ethanol to <30% compared to the control group. The black chokeberry red pigment fraction had a similarly significant protective effect on gastric mucosa in a dose-dependent manner when administered at 30-300 mg/kg of body weight, and the administration of 30 mg/kg of body weight could suppress ethanol-induced gastric mucosal damage by approximately 50% (ID(50) = 30 mg/kg of body weight). 相似文献

The effect of acetic acid on the dissolution of proteins in rice was studied to elucidate the mechanism for the textural change induced by the acid by chemical and SDS-PAGE analyses of the rice proteins in the soaking solution. More proteins were extracted with 0.2 M acetic acid (pH 2.7) than with water (pH 6.8). The effect of acetic acid on the protein dissolution increased with increasing temperature. Immunoblotting confirmed that, when rice was soaked in acetic acid, glutelin was dissolved into the soaking solution and degraded by aspartic proteinase. Aspartic proteinase degraded glutelin much more than it did albumin and globulin. It was found that the combined amount of albumin and globulin dissolved into the acetic acid solution was much larger than that of glutelin, despite the smaller amounts present of albumin and globulin than of glutelin. Metal ions were extracted more with acetic acid than with water. In addition, carboxypeptidase was activated under the acidic condition and resulted in an increase in the amount of free amino acids. The main effect of acetic acid on the dissolution of rice proteins was enhancement of the solubility of albumin, globulin, and glutelin, the effect of proteases being minor. 相似文献

Single cells prepared from autoclaved soybeans and cellulase treatment of the cells were effective in digesting the cell walls of and extracting the oil from soybeans. The first cell wall of the soybean single cell was completely removed using cellulases; the thin and transparent second cell wall of the cell was swollen. Oil in the cell formed spherical or hemispherical oil drops, and oil leaking from the oil bodies was observed. The oil was almost retained within the second cell wall. Water-extractable substances were obtained at approximately >60% of the weight. Flotation of oil drops by centrifugation was easily done. Ambient n-hexane extraction was also possible; however, residual oil remained in the oil bodies. Protease or peptidase digested the structure of the oil bodies; however, separation of the oil and the hydrolysates was impossible. The oil from the oil bodies was obtained effectively (>85%) by pressing the single cells and/or cellulase-treated single cells. 相似文献

Buffers,barriers and blankets: The (dis)connectivity of catchment-scale sediment cascades 总被引:2,自引:0,他引:2
Catchment configuration and the nature of connectivity within and between landscape compartments affect the operation of sediment cascades and geomorphic responses to disturbance events of differing magnitude and frequency. This paper introduces the concept of landform impediments, termed buffers, barriers and blankets, that impede sediment conveyance by limiting the connectivity between landscape compartments. Buffers restrict sediment delivery to channels, barriers inhibit sediment movement along channels, and blankets drape channel or floodplain surfaces affecting the accessibility of sediment to entrainment. These features operate as a series of switches which turn on/off processes of sediment delivery, determining the effective catchment area at any given time. Using previously documented examples, the role of these features in affecting the operation of sediment cascades in a low relief, passive landscape setting such as the Australian landmass is contrasted to examples from high relief, uplifting settings in New Zealand. The Australian examples are further explored by examining how changes to landscape connectivity brought about by human disturbance since European settlement have impacted upon landscape sensitivity and prospects for river recovery. This approach to analysis of impediments to sediment conveyance is generic and can be applied in any environmental setting. 相似文献

We studied the effects of TAK-044, a nonselective endothelin (ET) receptor antagonist, on the indomethacin- or methylene blue-induced constriction of the ductus arteriosus (DA) in rats and compared them with the effects on spontaneous DA constriction. Injection of TAK-044 into 21-day-old fetuses in utero was performed through the uterine wall of laparotomized mother rats under light ether anesthesia. The fetuses were autopsied 3 hr after treatment with TAK-044 (10 mg/kg) in utero and simultaneous administration to the laparotomized mother rats of indomethacin (3 mg/kg, p.o.) or methylene blue (100 mg/kg, i.p.). In the second experiment, pregnant rats were decapitated on day 21 of gestation to obtain newborn rats by cesarean delivery. Newborn rats which were given TAK-044 (2, 10 mg/kg) immediately after or 1 hr before cesarean delivery were autopsied at various times after birth. In both experiments, pups were rapidly frozen in an acetone-dry ice mixture at autopsy to evaluate the DA constriction by the whole-body freezing and shaving method. TAK-044 injection into the fetus 3 hr before autopsy completely inhibited the DA constriction induced by maternal treatment with indomethacin or methylene blue. TAK-044 caused dose-dependent inhibition of the spontaneous closure of the DA after birth. The inhibitory effect was more pronounced in pups which were given TAK-044 in utero 1 hr before birth; however, the inhibitory effect was incomplete in newborn pups. These results, together with the previous finding that BQ-123, an ETA-specific receptor antagonist, inhibits the ductal constriction induced by oxygen in vitro [Coceani et al., 1992], indicate that the ETA receptor plays a significant role in the indomethacin- or methylene blue-induced DA constriction as well as in the spontaneous DA constriction after birth, and also indicate that the inhibition of ETA receptor by TAK-044 was more easily achieved in fetuses than in neonates. 相似文献

An ELISA, using viable bovine lymphosarcoma cells, was developed to detect tumor-associated antigens (TAA) expressed on lymphosarcoma cells from cows with enzootic bovine leukosis (EBL). Monoclonal antibodies (MAB) against TAA were used. Using viable-cell ELISA, MAB reacted with tumor cells from 9 cows with EBL, but not with peripheral blood lymphocytes from 10 clinically normal cows. Titers of MAB against tumor cells from 1 cow with EBL were 2,048 (direct immunofluorescence assay), 8,192 (flow cytometry), 2,048 (fixed-cell ELISA), and 16,384 (viable-cell ELISA). Viable-cell ELISA was the most sensitive method for detection of TAA. Reactivities of 7 MAB to tumor cells from cows with EBL were compared between viable-cell ELISA and a complement-dependent antibody cytotoxicity test. Of 5 MAB with no cytotoxic activity against tumor cells, 3 were reactive against tumor cells from the same cow, as determined by viable-cell ELISA, with titers ranging from 128 to 2,048. 相似文献

To improve the cryopreservation protocol for mouse sperm, we attempted to estimate the type and extent of cryoinjury at various steps of the process. First, we demonstrated that mouse sperm are sensitive to chilling at -15 C and that the sensitivity is dependent on the length of exposure. To estimate cryoinjuries, sperm suspensions were ice-seeded at -5 or -15 C, frozen with liquid nitrogen (LN(2)) gas and then frozen in LN(2). In one experiment, sperm seeded at -5 C were cooled slowly to -15 C before deep freezing. At various steps of the cryopreservation process, the sperm were warmed and their viability was assessed based on motility and the integrities of the plasma membrane and acrosome. The motility of frozen-thawed sperm was higher on seeding at -5 C (28%) than at -15 C (9%). The motility did not decrease when the sample was transferred from LN(2) gas to LN(2). To estimate cryoinjury of sperm, we presumed the viability of frozen sperm to be decreased by chilling, hypertonic stress and formation of intracellular ice. When the sperm suspension was cooled and seeded at -5 C, the motility decreased by 25% due to hypertonic stress. When the sperm were cooled in LN(2) gas, the motility decreased by 17% with the formation of intracellular ice. When the sperm were cooled to -15 C, the motility decreased by 51% from chilling. After seeding, the motility decreased by 18% due to formation of intracellular ice and by 7% due to hypertonic stress. Considering the results, it would be preferable to seed samples at a higher temperature to prevent intracellular ice from forming and to cool seeded samples rapidly enough to minimize chilling injury and hypertonic stress, but not too rapidly to allow intracellular ice to form. 相似文献

Movement of water and cryoprotectants through the plasma membrane needs to be accelerated for successful cryopreservation of zebrafish oocytes/embryos, which are much larger than their mammalian counterparts. Aquaporin-3 is a water/solute channel that can transport not only water but also various cryoprotectants. In this study, we attempted to increase the permeability of immature zebrafish oocytes at stage III to water and cryoprotectants by exogenous expression of rat aquaporin-3. Immature zebrafish oocytes were injected with rat aquaporin-3 cRNA and cultured for 5-12 h. Permeability to water and cryoprotectants was then determined based on changes in the volumes of the oocytes in a hypertonic sucrose solution and various cryoprotectant solutions at 25 C. The permeability to water of the aquaporin-3 cRNA-injected oocytes was three times higher than that of intact and water-injected oocytes. The permeability of the aquaporin-3 cRNA-injected oocytes to ethylene glycol, glycerol, propylene glycol, and DMSO was also 2-4 times higher than that of intact oocytes. Thus, the permeability of immature zebrafish oocytes to water and cryoprotectants was enhanced by exogenous expression of aquaporin-3. Cryopreservation of teleost oocytes may be realized through a further increase in permeability. 相似文献
